I am rebuilding another motor (#4) for a Honda Hobbit PA50I. Last week I ordered a new reed for the motor from a Treats. However they sent me a reed for a PA50 II by mistake.
If I grind off the little nub on the reed and use a intake manifold from a PA50II will this combination work with the original PA50I carb and do I need to make any other changes?

I was going to say exactly that. If u r "rebuilding " follow the wiki to convert 501 to 502. But if u r putting 2 Reed's on a 1, I'm not really sure how that runs. Let us know please. I'm curious.
If you put pa50ii reeds on a pa50i reedblock your bike will run like total shit. If you replace the entire reedblock with the pa50ii reedblock you will just gain some speed/acceleration.
I think you mean that you have a pa50ii reedblock, which is fine, but if you just have the reeds on the pa50i reedblock you're in for a bad time.
Nick: Yes, I am installing a PA50II reed with a PA50II reed block on a PA50I motor with a PA50I carb. Are any adjustments to carb required like a larger main jet?

Before I got a PA50II intake, I ran the dual reeds on a PA50I intake and it ran fine,though no better - but find you a PA50II intake - way better/bigger.

Nah you'll be fine. If anything you might have to downjet but you should probably not notice anything. The pa50ii reeds are actually much better than the pa50ii so you might notice a slight performance increase.
Nick: I finally got around to installing the PA50II (reeds and intake) on the PA50I motor and have it running. But it just does not seem to run right. What size jet(s) should I try to improve the running. Thanks Jack
